The alignment of Route 151 originates to 1946, when the state took over jurisdiction of South Tenth Street, South Eleventh Street and Flanders Avenue from an entrance road on State Highway Route 25 near the Cooper River.

State Highway Route 39 continued northward, intersecting with U.S. Route 130 and State Highway Route 25 in Bordentown.
Prior to the 1953 renumbering, Route 413 was Route S25, a prefixed spur of New Jersey Route 25.
