Holmes underwent a clitorectomy, described as a "clitoral recession", at age 7, at The Hospital for Sick Children in Toronto.
He is a member of the Board of Trustees of the The Hospital for Sick Children, a member of the Advisory Board for the Richard Ivey School of Business, a member of the Board of Directors of the Royal Ontario Museum Foundation and as well as Chairman of the Investment Committee of St. Paul’s Anglican Church.

The hospital was originally completed at Garnethill in 1882 and opened on 20 December as the Hospital for Sick Children.
Prior to his appointment as the Vice-Chancellor, he was Geneticist-in-Chief and Head of the Genetics and Genomic Biology Program of the Research Institute at the Hospital for Sick Children and co-founder (with Dr. Steve Scherer) of The Centre for Applied Genomics.
