He was ordained a Catholic Priest in 1728, and made a Cardinal in 1733, given the titulus of San Cesareo in Palatio.

On February 5, 2011, the memorial of St. Agatha, Cardinal Burke took canonical possession of his titular church in Rome, Sant'Agata dei Goti.
It was rebuilt under Pope Nicholas V, had its long-held titular-church status suppressed by Pope Sixtus V, was renovated by Francesco Barberini in 1643, and rebuilt and given to the Society of the Sacred Heart of Jesus by Pope Clement XI and his architect Carlo Fontana in 1703-1705.
The present Cardinal Priest of this Titular Church is Joseph Zen Ze-kiun, SDB (Traditional Chinese: 陳日君樞機), the Diocesan Bishop of the Diocese of Hong Kong in China.
