Soon after his enthronement he forwarded his noted synodical letter to Pope Honorius I and to the Eastern patriarchs, explaining the orthodox belief in the two natures, human and divine, of Christ, as opposed to Monothelitism, which he viewed as a subtle form of heretical Monophysitism (which posited a single divine nature for Christ).

In 637, after a long siege of Jerusalem, Patriarch Sophronius surrendered Jerusalem to Caliph Umar, but secured the Covenant of Umar I, which recognised Christian rights to protection.
The Greek tradition has a long unbroken history of elders and disciples, such as Sophronius and John Moschos in the seventh century, Symeon the Elder and Symeon the New Theologian in the eleventh century, and contemporary charismatic gerontes such as Porphyrios and Paisios.
Later, Saints Epiphanius of Salamis, Gregory of Tours, Isidore of Seville, Modest, Sophronius of Jerusalem, German of Constantinople, Andrew of Crete, and John of Damascus talk about the tomb being in Jerusalem, and bear witness that this tradition was accepted by all the Churches of East and West.
