He, along with Diego de Mazariegos, founded the City of “Villareal de Chiapa de los Españoles”, now San Cristobal de las Casas, Chiapas, Mexico, in 1528 as a regional base for the conquest of Guatemala.

After several attempts to conquer Guatemala, in 1525, Hernán Cortés sent Diego de Mazariegos and Andrés de la Tovilla, along with a group of 150 foot soldiers and forty horses, to complete the conquest.
In 1570 when Diego de Mazariegos took charge as governor of the province of Venezuela, he made it a priority to pacify the territories.
