La Gloria is situated to the Eastern margin of the Magdalena River, it borders the north with the municipalities of Tamalameque and Pelaya, the south with the municipalities of Gamarra and Aguachica, the West with the Bolívar Department and the east with the Norte de Santander Department.
It is connected to Perote by a small highway; there, the small highway intersects Federal Highway 140, which goes to Xalapa.
Controversy also surrounds the half-owned Smithfield Foods pig sheds near the village of La Gloria, Mexico, where as many as 1,800 villagers living near the plant had already complained of respiratory problems and 400 had been treated before the 2009 swine flu outbreak.

It is bounded on the North by J. J. Cartagena Street (Las Delicias community), Ruth Fernandez Boulevard, and Camino de Ponce (Golf Club) Road, on the South by PR-123, Ausencia Street, and La Gloria Street, on the West by the hills west of PR-123, and on the East by Novedades Street, the hills east of Ponce Cement and the future western branch of PR-9.
In Michelangelo Antonioni's film, The Passenger, the character of Locke (played by Jack Nicholson) is assassinated in a hotel located in Osuna (Hotel de la Gloria).
