The Province was named for its most important part, Isla Margarita.

He then became Governor of the Margarita Province, based on Isla Margarita off the coast of what today is Venezuela, from 1630 to 1638 before retiring to Spain.
Early on, the province was defined in relation to the Venezuelan coastline (with Margarita Province to the north covering the Isla Margarita region).
